Background. High-Performance Thin-Layer Chromatography (HPTLC) is a simple, flexible and cost efficient method for quantitative analysis. Analysis can be performed easily in short time and requires small amount of the sample. Silene wallichiana (Caryophyllaceae) have been previously shown to contain 20-hydroxyecdysone (20E) and recognized as a perspective source of ecdysteroids. From the literature survey and to our best knowledge no previous report is available on determination and quantification of 20E in Silene species using HPTLC. Purpose. The aim of the work is to develop a simple, rapid, selective and cost effective HPTLC method for the determination and quantification of 20E in S. wallichina. Methodology. HPTLC method was developed for the estimation of 20E in S. wallichina. The method employed TLC aluminum plate precoated with silica gel 60F254 as the stationary phase. The solvent system employed consisted of THF / toluene / 1mM TFA in MeOH / H2O (16:8:2:1, v). Quantitative analysis was performed by densitometric scanning at 249 nm. Originality. This is the first time that an HPTLC densitometric method was applied to quantify the amount of the 20E in the methanol extract of S. wallichiana. Findings. We are the first to report the HPTLC analysis of methanol extract S. wallichina. The plant material was extracted with methanol and analyzed by HPTLC for 20E content. Results of HPTLC analysis showed that the content of 20E was 131.6 µg/mg (dry mass) in S. wallichiana. Highlights: - methanol extract of Silene wallichiana; - quantity of 20-hydroxyecdysone in Silene wallichiana; - HPTLC method development for the quantification of 20-hydroxyecdysone
